| Description | Prepared by separating erythrocytes from plasma by centrifugation. The plasma is siphoned off and ether is added to the erythrocyte paste causing the cells to lyse. A second centrifugation removes the cell walls resulting in a clear red solution of hemoglobin. Hemoglobin is a tetramer (which contains 4 subunit proteins) called hemoglobin A, consisting of two alpha and two beta subunits non-covalently bound, each made of 141 and 146 amino acid residues, respectively. The subunits are structurally similar and about the same size. Each subunit has a molecular weight of ~17kD, for a total molecular weight of the tetramer of ~68kD. Recommended Dilution: Optimal dilutions to be determined by the researcher. |
