| Description | Purity ~95% (SDS-PAGE, HPLC). Proneurotrophin-3 (ProNT-3) is the precursor form of neurotrophin 3. ProNT3 functions via p75NTR and sortilin and induces neuronal apoptosis. On the contrary, mature NT3 interacts with Trk receptors and selectively promotes the survival, growth and differentiation of neurons. As axon retrograde transport is crucial for neurotrophin signaling, Pro-NT3 also plays an important role in the mechanism of retrograde signaling. ProNT3 interacts preferentially with the pan-neurotrophin receptor p75 (p75NTR) and vps10p domain-containing receptor sortilin and induces neuronal apoptosis, whereas mature BDNF selectively binds with high affinity to the TrkB kinase receptor and promotes the survival, growth and differentiation of neurons. As proneurotrophins and mature neurotrophins elicit opposite biological effects, Pro-BDNF cleavage in the neuronal system is regulated in a specific and cell-context dependent manner.
