| Description | Purity ~95% (SDS-PAGE). Flagellin is the subunit protein which polymerizes to form the filaments of bacterial flagella. It activates the innate immune system through the receptor Toll-like Receptor 5 (TLR5) or the intracellular NLRC4 protein. The Flagellin (NLRC4 Mutant)(rec.) is only detected by TLR5 not by NLRC4. Source: Recombinant protein corresponding to the native flagellin [FliC] from Salmonella enterica Serovar Typhimurium strain ATCC14028, aa1-495, contains a small deletion at the C-terminus of the protein required for NLRC4 activation, fused to His-tag at N-terminal, expressed in E. coli. Biological Activity: Activation of TLR5 in human epithelial cell assays based on NF-kB luciferase fusions, in vivo on expression of CCL20 or IL-6. TLR5 can be stimulated in vitro by Flagellin (NLRC4 mutant) at a concentration range of 5-50ng/ml and in vivo at a starting concentration of 0.1-1ug/mouse. Does not activate NLRC4 as tested in bone-marrow mouse macrophages. It can be used as negat |
