MachOne three-dimensional multifunctional micromechanical testing system is the only integrated micromechanical testing device that can integrate compression, tension, shear, friction, torsion and indentation testing. It can provide precise and reliable mechanical stimulation and characterization for biological tissues, polymers, gels, biomaterials, capsules, adhesives and foods. The mechanical properties allowed to be characterized include stiffness, strength, modulus, viscoelasticity, plasticity, hardness, adhesion, swelling and relaxation, displacement control motion.
INDENTATION AND THICKNESS MAPPING ON SAMPLES OF DIFFERENT STIFFNESSES IN A 96-WELL PLATEThe testing of biological materials as well as bacteriological, viral, and cell cultures is made easier with well plates. They contain many plastic-formed wells that are used as miniature test tubes. This is excellent for downsizing test space and for performing many tests at once. In this case study, a multiaxial mechanical tester model MachOne v500css was used to determine the thickness and stiffness of small samples of agarose gels in a 96-well plate. Multiple concentrations were tested from 0.8% to 1.2% and with volumes of these gels varying from 0.1 to 0.5 mL. The samples were made from 6 mm biopsy punched disks instead of pouring the Agar solution directly into the wells in order to ensure a flat surface for indentation testing.
By implementing a custom script on the obtained data, the thickness and indentation modulus of each sample was extracted using Hayes Elastic Model. The results obtained show that the modulus is higher for samples of higher concentration of agar. Further analyses can be performed by modifying the script or performing statistical analyses on the data.
